Making a Div Layout from scratch?

I wanted to take this image

and make it the background of a div profile and place boxes over the appropriate sections...

How would i do this i have tried doing it myself but cant get it right so i came here.

Use a basic DIV code:

Then place the right nmbers into left and top pixels, until you have it right. Make a guess and then fine tune it. With practice, you can look at an area and guess the right amount of pixels. What server are you using?
